KINETICS OF FREE CARBONIC ANHYDRASE IN CONVERSION OF p-NITROPHENYL ACETATE INTO p-NITROPHENOL

Carbon capture by using carbonic anhydrase becomes one of the alternative environmental friendly approaches nowadays. Carbonic anhydrase (CA) is naturally found enzyme in human blood and other mammals as transportation aid for carbon dioxide (CO2) during respiration process. İn this study, the characterization of CA kinetics were determined by using p-NPA as a substrate at 25ºC. The concentration of the p-NPA was varied from 0.5-2.5mM meanwhile the enzyme concentration was kept constant. The kinetics parameters were determined by using Lineweaver-Burk plot. The Km, Vm and kcat value of free CA enzyme were 14.29mM, 0.193µmol/ml/min and 1.93s 1respectively. Based on the value obtained, it indicated that complex substrate-enzyme was well bind for catalyzing the hydrolysis reaction of p-NPA to p-NP product.
